Generic Name: Emflaza
What is Emflaza?
Temazepam: The CNS-depressant effects of MAOIs can be potentiated with concomitant administration of other drugs known to cause CNS depression including benzodiazepines. Terazosin: Additive hypotensive effects may be seen when monoamine oxidase inhibitors MAOIs are combined with antihypertensives. Teriflunomide: Use caution when administering teriflunomide with selegiline.
In vivo data suggest that teriflunomide is a weak inducer of CYP1A2. Coadministration of teriflunomide with CYP1A2 substrates, such as selegiline, may decrease selegiline exposure and lead to efficacy reduction. If teriflunomide is discontinued in a patient taking selegiline, serum concentrations may increase. Monitor patients for increases in adverse effects, such as dyskinesia, hallucinations, or nausea.
Dose adjustments may be required. Tetrabenazine: Tetrabenazine depletes presynaptic dopamine, norepinephrine, and serotonin storage and antagonizes postsynaptic dopamine receptors.
Therefore, concurrent use of tetrabenazine with monoamine oxidase inhibitors is contraindicated.

What should I avoid while taking Emflaza?
Hypotension, sometimes sudden in onset, has been reported with conventional selegiline. Special care should be taken when administering selegiline to patients who have labile hypertension, cardiac arrhythmias, severe angina pectoris, psychosis or a history of peptic ulceration as aggravation of these conditions may occur during treatment. Although serious hepatic toxicity has not been observed, caution is recommended in patients with a history of hepatic dysfunction.
Transient or continuing abnormalities with a tendency for elevated plasma concentrations of liver enzymes have been described during long-term therapy with conventional tablets of selegiline. MAO inhibitors, including selegiline, may potentiate the effects of CNS depressants used for general anaesthesia.
Emflaza side effects
Some studies concluded in an increased risk of mortality in patients receiving selegiline and levodopa compared to those receiving levodopa only. Studies have related the risk of an increased hypotensive response to concomitant administration of selegiline and levodopa, in patients with cardiovascular risk.
The addition of selegiline to levodopa may not be beneficial in those patients who experience fluctuations in response which are not dose dependent.
Caution is advised when selegiline is taken in combination with other centrally acting medicinal products and substances. The concomitant intake of alcohol should be avoided.
Pethidine The concomitant administration of the selective MAO-B inhibitor selegiline and pethidine is contraindicated. Selective serotonin reuptake inhibitors SSRIs and serotonin noradrenaline reuptake inhibitors SNRIs Because of the risk of confusion, hypomania, hallucination and manic episodes, agitation, myoclonus, hyperreflexia, incoordination, shivering, tremor, convulsion, ataxia, diaphoresis, diarrhea, fever, hypertension, which can be part of the serotonine syndrome, concomitant administration of selegiline and SSRIs or SNRIs is contraindicated.
Common Emflaza ide effects may include:
Selegiline did not induce mutations or chromosomal damage when tested in the bacterial mutation assay in Salmonella typhimurium and in an in vivo chromosomal aberration assay.
Caution patients against rising rapidly after sitting or lying down, especially if they have been doing so for prolonged periods and especially during treatment initiation.
Spironolactone: Additive hypotensive effects may be seen when monoamine oxidase inhibitors MAOIs are combined with antihypertensives.
Amiloride: Additive hypotensive effects may be seen when monoamine oxidase inhibitors MAOIs are combined with antihypertensives.
