What is Colcrys?

Additional pharmacological actions of clozapine that further help to reduce depression and anxiety have also been reported in the literature. Clozapine is used for the control of schizophrenia in the patients who have failed to respond to any other antipsychotic agent.

In schizophrenia patients, clozapine has also been evidenced to decrease the tendency for substance abuse. This beneficial effect of the drug is related to its weak binding to dopamine receptor as compared to other antipsychotics. Clozapine is associated with a number of adverse effects some of which are potentially life-threatening. Following is a brief description of the possible risks:Different isomers of liver cytochrome P450 enzymes have been identified that metabolize clozapine.

Whereas CYP1A2 is suggested to be the main enzyme that metabolizes clozapine, the contribution of CYP2C19 is moderate and CYP3A4 is functional in the patients wherein CYP1A2 has been compromised.

Polymorphism among these enzymes leading to the existence of a number of genetic variants, both active and inactive is also known. The individuals, homozygous for the inactive variants metabolize the drug poorly, whereas those homozygous with normal or active variants can metabolize clozapine efficiently.

The two kinds of people will require different doses to obtain the same effect that is more beneficial and less toxic. In addition, polymorphism of ABCB1, a protein responsible for transporting clozapine across plasma membrane has also been reported to affect the blood concentrations of clozapine. In order to determine the type of enzyme variants present in an individual, genetic testing is required.

Could this medication cause this. A: Clozapine has many serious side effects that could occur, and this is the reason why you are monitored closely and have to go for bloodwork so often while taking this medication. If you are experiencing any type of problems related to heart rate you should tell your doctor about it as soon as possible because this is not a normal side effect of the medication. This type of side effect can be serious if left unchecked, so it is best to call your physician.

Burton Dunaway, PharmDQ: Can taking Clozaril raise your potassium level. A: Clozaril clozapine is an atypical antipsychotic medication. Clozaril is approved for use in treatment-resistant schizophrenia. Clozaril is also indicated for reducing the risk of recurrent suicidal behavior in people with schizophrenia or schizoaffective disorder. Clozaril works by changing chemicals in the brain.

According to the prescribing information, a increase in potassium levels was not a reported side effect of the medication. Clozaril may lead to agranulocytosis. Agranulocytosis is a very serious blood condition when the body fails to produce enough white blood cells. A doctor or health care provider should be contacted if fever, sore throat, chills, weakness, tiredness, or other signs of flu are experienced.

To decrease this risk it is recommended that the blood be regularly monitored. Common side effects include constipation, bed-wetting, night-time drooling, muscle stiffness, sedation, tremors, orthostatic hypotension, hyperglycemia, and weight gain.

What should I avoid while taking Colcrys?

Hyperglycemia, in some cases extreme and associated with ketoacidosis or hyperosmolar coma or death, has been reported in patients treated with atypical antipsychotics including CLOZARIL. Assessment of the relationship between atypical antipsychotic use and glucose abnormalities is complicated by the possibility of an increased background risk of diabetes mellitus in patients with schizophrenia and the increasing incidence of diabetes mellitus in the general population.

Given these confounders, the relationship between atypical antipsychotic use and hyperglycemia-related adverse reactions is not completely understood. However, epidemiological studies suggest an increased risk of treatment-emergent, hyperglycemia-related adverse reactions in patients treated with the atypical antipsychotics. Precise risk estimates for hyperglycemia-related adverse reactions in patients treated with atypical antipsychotics are not available. Patients with an established diagnosis of diabetes mellitus who are started on CLOZARIL should be monitored regularly for worsening of glucose control.

Colcrys side effects

It showed to be most useful in refractory manic disorders. They randomly assigned 262 bipolar disorder patients in a short term manic or mixed episode to abilify monotherapy or placebo. Patients were admitted for minimum of two weeks and YMRS was used as a measuring scale. This scale showed a statistical improvement in bipolar symptoms with patients on abilify compared to placebo -8.

Adverse effects were not a significant issue, but completion of the study was an issue with a few patients. In addition, Abilify was well tolerated in the study discussed above. Because they are both effective as monotherapies, the patient may be administered an appropriate monotherapy dose of Clozaril for refractory bipolar disorder.

Effectiveness and side effects should be closely monitored.
