Question: What is buspirone Busparhow does it work, and how is it used to treat anxiety disorders. Answer: Buspirone, also known by its trade name Buspar, is a non-benzodiazepine medication that is indicated for the treatment of generalized anxiety disorder. It's thought to work through its binding and activation of a certain type of serotonin receptor in the brain -- specifically the 5HT1A receptor -- and it's through this activity that buspirone is thought to have its anti-anxiety effects.
When buspirone first came on the market it was shown in clinical trials to have effectiveness equal to benzodiazepines in the treatment of generalized anxiety disorder without the sedation that frequently comes with benzodiazepines, so it was a much heralded drug when it first came out.
The drug, sold under the brand name Buspar by the Mead Johnson Pharmaceutical division of the Bristol-Myers Company, was shown in clinical trials to be as effective for treating anxiety as benzodiazepines, the most popular existing tranquilizers, but without some of the more dangerous side effects. Most notably, Buspar caused far less drowsiness than other tranquilizers and did not produce physical dependency in patients after prolonged use, a major drawback of Valium and other benzodiazepines, which have been blamed for severe withdrawal symptoms in some patients when therapy was abruptly discontinued.
Buspar pronounced BYEW-spar was also deemed safer to take in conjunction with alcohol because it did not exacerbate the effects of alcohol as the benzodiazepines did. It also appeared less likely to be overused because it did not give the patients a euphoric high or buzz.
How should I take Buspar?
Buspirone hydrochloride is a white crystalline, water soluble compound with a molecular weight of 422. The 30 mg tablet also contains iron oxide. The mechanism of action of buspirone is unknown. Buspirone differs from typical benzodiazepine anxiolytics in that it does not exert anticonvulsant or muscle relaxant effects. It also lacks the prominent sedative effect that is associated with more typical anxiolytics. In vitro preclinical studies have shown that buspirone has a high affinity for serotonin 5-HT1A receptors.
Buspirone has no significant affinity for benzodiazepine receptors and does not affect GABA binding in vitro or in vivo when tested in preclinical models.
