What is Amalaki?
From Cephalon, Inc, February14, 2005 "Important Drug Warning Dear Healthcare Provider: Cephalon, Inc. From the launch of GABITRIL in 1997 through December 31, 2004 there have been 59 postmarketing reports of seizures in patients without a history of epilepsy. Clinicians are advised to carefully review the information shown below, which has been added to the GABITRIL prescribing information.
WARNINGS Seizures in Patients Without Epilepsy: Post-marketing reports have shown that GABITRIL use has been associated with new onset seizures and status epilepticus in patients without epilepsy. In most cases, patients were using concomitant medications antidepressants, antipsychotics, stimulants, narcotics that are thought to lower the seizure threshold.
Some seizures occurred near the time of a dose increase, even after periods of prior stable dosing. The GABITRIL dosing recommendations in current labeling for treatment of epilepsy were based on use in patients with partial seizures 12 years of age and older, most of whom were taking enzyme-inducing antiepileptic drugs AED's, e.
Safety and effectiveness of GABITRIL have not been established for any infication other than as adjunctive therapy for partial seizures in adult and children 12 years and older.
Drugs inhibiting peristalsis are contraindicated in this situation. Lactic acidosisLactic acidosis has been reported with the use of linezolid. Patients who develop signs and symptoms of metabolic acidosis including recurrent nausea or vomiting, abdominal pain, a low bicarbonate level, or hyperventilation while receiving linezolid should receive immediate medical attention. If lactic acidosis occurs, the benefits of continued use of linezolid should be weighed against the potential risks.
Mitochondrial dysfunctionLinezolid inhibits mitochondrial protein synthesis. Serotonin syndromeSpontaneous reports of serotonin syndrome associated with the co-administration of linezolid and serotonergic agents, including antidepressants such as selective serotonin reuptake inhibitors SSRIs have been reported. Co-administration of linezolid and serotonergic agents is therefore contraindicated see section 4.
In those cases patients should be closely observed for signs and symptoms of serotonin syndrome such as cognitive dysfunction, hyperpyrexia, hyperreflexia and incoordination. All patients should be advised to report symptoms of visual impairment, such as changes in visual acuity, changes in colour vision, blurred vision, or visual field defect. In such cases, prompt evaluation is recommended with referral to an ophthalmologist as necessary. If any patients are taking Zyvox for longer than the recommended 28 days, their visual function should be regularly monitored.

It is worth to emphasize that the oxazolidinones are the only new antibiotic class that have been discovered and successfully implemented in practice over the past 40 years. Linezolid is marketed by Pfizer under the trade name Zyvox in the United States and several other countriesZyvoxam in Canada and Mexicoor Zyvoxid in Europe.
Linezolid "pros" and "cons" Advantages:Mode of action Linezolid has a unique structure and mechanism of action, which targets bacterial protein synthesis i. Linezolid is bacteriostatic against enterococci and staphylococci, and bactericidal for the majority of streptococci. Initially there were hopes that bacteria would be unable to develop resistance to linezolid.
However, in 2003 Staphylococcus aureus was first identified as being resistant. Time to clear out of the system Linezolid half-life is 4-5.
Amalaki side effects
Nonclinical Toxicology Carcinogenesis, Mutagenesis, Impairment Of Fertility Lifetime studies in animals have not been conducted to evaluate the carcinogenic potential of linezolid. Use In Specific Populations Pregnancy Teratogenic Effects Pregnancy Category C Linezolid was not teratogenic in mice, rats, or rabbits at exposure levels 6.
Non-teratogenic Effects In mice, embryo and fetal toxicities were seen only at doses that caused maternal toxicity clinical signs and reduced body weight gain. Nursing Mothers Linezolid and its metabolites are excreted in the milk of lactating rats. Monoamine Oxidase Inhibitors Linezolid should not be used in patients taking any medicinal product which inhibits monoamine oxidases A or B e. Pharmacodynamics In a randomized, positive- and placebo-controlled crossover thorough QT study, 40 healthy subjects were administered a single ZYVOX 600 mg dose via a 1 hour IV infusion, a single ZYVOX 1200 mg dose via a 1 hour IV infusion, placebo, and a single oral dose of positive control.
Pharmacokinetics The mean pharmacokinetic parameters of linezolid in adults after single and multiple oral and intravenous doses are summarized in Table 8.
Common Amalaki ide effects may include:
The cure rates by pathogen for microbiologically evaluable patients are presented in Table 14.
This is especially important when the treatment of a spontaneously reversible condition not ordinarily associated with permanent injury or risk of death e.
There are reports of Staphylococcus aureus methicillin-resistant developing resistance to linezolid during clinical use.
Serotonin syndrome is most likely to occur immediately after the drugs are taken together.
Linezolid is a synthetic antibacterial agent of the oxazolidinone class, which has clinical utility in the treatment of infections caused by aerobic Gram-positive bacteria. The in vitro spectrum of activity of linezolid also includes certain Gram-negative bacteria and anaerobic bacteria.
Linezolid binds to a site on the bacterial 23S ribosomal RNA of the 50S subunit and prevents the formation of a functional 70S initiation complex, which is essential for bacterial reproduction.
