What is Addyi?

The safety and efficacy of nifedipine in children below 18 years have not been established. Currently available data for the use of nifedipine in hypertension are described in section 5. The pharmacokinetics of Adalat capsules are altered in the elderly so that lower maintenance doses of nifedipine may be required. In patients with mild, moderate or severe impaired liver function, careful monitoring and a dose reduction may be necessary.

The safety of Adalat 10mg during pregnancy or in nursing mothers has not been established see sections 4. Adalat 10 mg must not be used in cases of cardiogenic shock, clinically significant aortic stenosis, unstable angina pectoris, or during or within 4 weeks of an acute myocardial infarction.

Adalat 10 mg must not be administered concomitantly with rifampicin since effective plasma levels of nifedipine may not be achieved owing to enzyme induction See Section 4. Adalat 10 mg may be used in combination with beta-blocking drugs and other antihypertensive agents but the possibility of an additive effect resulting in postural hypotension should be borne in mind.

Adalat 10 mg will not prevent possible rebound effects after cessation of other antihypertensive therapy. Care must be exercised in patients with very low blood pressure severe hypotension with systolic pressure less than 90 mm HGin cases of manifest heart failure and in the case of severe aortic stenosis.

Treatment with short-acting nifedipine may induce an exaggerated fall in blood pressure and reflex tachycardia, which can cause cardiovascular complications such as myocardial and cerebrovascular ischaemia. As with other vasoactive substances, angina pectoris may very rarely occur data from spontaneous reports with immediate release nifedipine, especially at the start of the treatment.

Data from clinical studies confirm that the occurrence of angina pectoris attacks is uncommon.

How should I take Addyi?

The parameters for preload, central venous pressure and pulmonary capillary wedge pressure did not show any statistically significant changes. What should I avoid while taking Addyi?

Clopidogrel: No clinically significant pharmacodynamic interactions were observed when clopidrogrel was co-administered with nifedipine.

Tirofiban: Co-administration of nifedipine did not alter the exposure to tirofiban importantly. Diuretics, PDE5 inhibitors, alpha-methyldopa: Nifedipine may increase the blood pressure lowering effect of these concomitantly administered agents. Ketoconazole, itraconazole and fluconazole are CYP3A inhibitors and can inhibit the metabolism of nifedipine and increase the exposure to nifedipine during concomitant therapy. Blood pressure should be monitored and a dose reduction of nifedipine considered.

Omeprazole: In healthy volunteers receiving a single dose of 10 mg nifedipine, AUC and Cmax of nifedipine after pretreatment with omeprazole 20 mg q.

Addyi side effects

Atenolol 50 mg once daily was used concomitantly in 187 of the 370 patients on Adalat CC and in 64 of the 126 patients on placebo. All adverse events reported during Adalat CC therapy were tabulated independently of their causal relationship to medication. The most common adverse event reported with Adalat CC was peripheral edema. Nifedipine is mainly eliminated by metabolism and is a substrate of CYP3A. Inhibitors and inducers of CYP3A can impact the exposure to nifedipine and consequently its desirable and undesirable effects.

  • If you have questions about the drugs you are taking, check with your doctor, nurse or pharmacist.

  • Inform your doctor of any medical conditions you may have or that you have experienced in the past including heart failure, low blood pressure, liver disease, diabetes of kidney disease.

Special considerations If you have liver disease, body may have trouble clearing nifedipine from your blood. Keep these considerations in mind if your doctor prescribes nifedipine oral tablet for you. General Adalat CC should be taken on an empty stomach.

Other versions of the drug can be taken either with food or on an empty stomach.

Where can I get more information?

  • Hemodynamic studies of the immediate release nifedipine formulation in patients with normal ventricular function have generally found a small increase in cardiac index without major effects on ejection fraction, left ventricular end-diastolic pressure LVEDP or volume LVEDV.

  • Swallow whole, do not chew or crush the capsule or its contents.
